This test analyzes a tumor and helps predict if the cancer will return, also known as the cancer’s recurrence rate, as well as the risk of metastasis (cancer spreading to other areas) and more for patients with stage I–III cutaneous melanoma. How Does Melanoma Genetic Testing Work?

Melanoma genetic testing from Castle Biosciences is a gene expression profile (GEP) test that looks at the activity of 31 genes in a melanoma tumor. By analyzing the tumor biology at the molecular level, the test gives a personalized risk score alongside the traditional AJCC staging system.

Along with the risk score, the test also gives a risk of recurrence as risk of cancer being found in your sentinel lymph node. Overall, this means that melanoma genetic testing gives three different results, which gives a comprehensive risk score and analysis.

What To Expect With Melanoma Genetic Testing

If you are newly diagnosed with melanoma, Dr. Chang will evaluate your tumor and determine if melanoma genetic testing is the right fit for you. If so, we will:

  • Take A Tumor Sample: The test uses tissue already removed during your biopsy or surgery—so you don’t have to worry about getting an extra procedure.

  • Send To The Lab: We will send the sample to Castle Biosciences’ specialized laboratory.

  • Review Results: Once we have received your results in about one to two weeks, Dr. Chang will review them with you and use the information to tailor your care plan and to help guide you in your next steps.

How Does The Test Work?

First, we will use your tissue for the test. Next, you can expect:

  • RNA extraction & cDNA synthesis: The lab isolates RNA, converts it to cDNA, and amplifies 31 target genes via RT‑PCR.

  • Algorithmic Analysis: The gene expression signature is run through a validated algorithm, integrating tumor biology and clinical‑pathologic factors.

  • Results: You will receive a comprehensive report that shows your class score or risk, your i31‑SLNB or your personalized risk of positive sentinel lymph nodes, and your i31‑ROR—your personalized risk of recurrence/metastasis.
